A full recording of last week’s webinar, “Transcreener Assays for Screening and Profiling GTPases” is now available.
GTPases, including the Ras/Rho enzymes and Gα proteins associated with GPCRs, are a large family of enzymes that act as binary switches for diverse cellular processes. They are generally active in the GTP-bound state and inactive following hydrolysis until GDP is again displaced by GTP. GTP hydrolysis and GTP/GDP exchange are in turn regulated by a diverse collection of GTPase accelerating proteins (GAPS) and GTPase exchange factors (GEFs). GTPases historically have been challenging HTS targets because assay methods rely on phosphate detection or enzyme-coupled assays, which are both prone to high background and/or false positives.
The webinar covered:
- How the Transcreener GDP Assays enable robust screening and profiling of GTPases using direct immunodetection of GDP with fluorescence polarization (FP), fluorescence intensity (FI) or TR-FRET readouts.
- The use of Transcreener for detection of small GTPases in the Ras/Rho family and Gα proteins and their associated GAPs as well as more challenging targets such as the GTPase domain of LRRK2.
- The Transcreener GDP assays are the only method that allows direct detection of GDP without the use of coupling enzymes.
