Recombinant VPS4A Enzyme, Human

Active, human VPS4A (AA 111-437) protein expressed and purified from Baculovirus-infected insect cells with an N-Terminal His tag. The enzyme has been thoroughly validated with the Transcreener ADP Assay Kit. For specific activity, please refer to the Certificate of Analysis for each individual enzyme lot. Purity is 85% in a composition of 20 mM HEPES, 500 mM NaCl, 1 mM TCEP, and 10% Glycerol (pH 7.6). Currently used with the Transcreener assay at a concentration of 270 nM in a 10 μL reaction.

Active VPS4A Enzyme – Ready for Inhibitor Discovery

Here, we use 2 µM ADP. The enzyme buffer included 50 mM HEPES (pH 7.5), 10 mM MgCl₂, 50 mM KCl, and 0.01% Triton X-100. The enzyme reaction took place for 120 minutes at 37°C. The ADP Detection Mix included 1X Stop & Detect Buffer B, 8 nM ADP² Alexa Fluor 633 Tracer, and 3.16 µg/mL ADP antibody. The Detection Mix was added, incubated for 60 minutes at room temperature, and read on a plate reader.
