Recombinant PLK1 Enzyme, Human

Recombinant human PLK1 (amino acids 1-603), N-terminal 6xHis tag, 69.1 kDa; expressed and purified from baculovirus-infected insect cells. The enzyme has been thoroughly validated with the Transcreener ADP² Kinase Assay Kits. For specific activity, please refer to the Certificate of Analysis for each individual enzyme lot. Purity is >85% in a composition of 50 mM Tris-HCl pH 8.0, 500 mM NaCl, 20% glycerol. Currently, PLK1 enzyme is used with the Transcreener assay at a concentration between 1-2 nM in a 10 μL reaction.

Active PLK1 Enzyme – Ready for Inhibitor Discovery

The assay demonstrates linearity when raw data is converted to ADP using a standard curve. We used 50 mM Tris (pH 8.0), 2 mM MgCl₂, 0.01% Triton X-100, 1 mM MnCl_2,and 1 mM DTT for the buffer, and ran with 15 µM PLKtide and 5 µM ATP at 37°C for 1 hour. The Detection Mix was added and incubated at room temperature for 60 minutes and read with a CLARIOstar plate reader.
