Recombinant IDH1 R132H Enzyme, Human

Active, human IDH1 R132H enzyme, full-length, N-terminal 6xHis-tagged protein expressed and purified from E.Coli. The enzyme has been thoroughly validated with the Hydriscreener NADPH Assay Kits. For specific activity, please refer to the Certificate of Analysis for each individual enzyme lot. Purity is >95% in a composition of 50 mM Tris-HCl pH 7.5, 100 mM NaCl, 20% glycerol, 1 mM TCEP. Currently, mutant IDH1 R132H enzyme is used with the Hydriscreener Assay at a concentration of 8 nM in a 10 μL reaction.

The assay demonstrates linearity when raw data is converted to NADPH using a standard curve. We used 25 mM Tris (pH 7.5), 10 mM MgCl₂, 150 mM NaCl, and 0.03% BSA for the buffer, and ran with 1 mM α-ketoglutarate (α-KG), and 20 µM NADPH at 30°C for 1 hour. The Detection Mix was added and incubated at room temperature for 60 minutes and read with a CLARIOstar plate reader.
