Active, human SMARCAL1 (AA 226-870) protein expressed and purified from baculovirus-infected insect cells with an N-Terminal 6xHis tag. The enzyme has been thoroughly validated with the Transcreener ADP² Assay Kit. For specific activity, please refer to the Certificate of Analysis for each individual enzyme lot. Purity is 95% in a composition of 50 mM HEPES, 100 mM NaCl, 10% Glycerol, 1 mM TCEP, pH 7.5.

Active SMARCAL1 Enzyme – Ready for Inhibitor Discovery

The assay demonstrates linearity when raw data is converted to ADP using a standard curve. We used 50 mM Tris pH 8.0, 10 mM MgCl₂, 0.01% Triton, and 1 mM DTT at 30°C for the 1-hour enzyme reaction. The Detection Mix was added and incubated at room temperature for 60 minutes and read with a CLARIOstar plate reader. Sheared salmon sperm DNA or WRN-H substrate DNA may be used for measuring forked DNA dependent ATPase activity.
